EasySep Dir HLAXM T Cell Iso Kit

From STEMCELL Technologies, this immunomagnetic cell isolation kit enables negative isolation of untouched human T cells directly from whole blood, spleen, or lymph node samples, without density gradient centrifugation or red blood cell lysis. The isolated cells are intended for serology or flow cytometry crossmatch assays. According to the manufacturer, starting with human whole blood from normal healthy donors, the T cell (CD3+) content of the non-lysed final isolated fraction typically ranges from 89 - 99.9% (gated on CD45), with a further manufacturer figure stating it as 97 ± 3% (gated on CD45). In a representative example, the T cell (CD3+) content of the lysed whole blood start sample and non-lysed final isolated fraction is 38.0% and 93.2% (gated on CD45), respectively.

The procedure uses column-free immunomagnetic separation and can also be applied to HetaSep™-treated whole blood, washed leukapheresis samples, and buffy coat. Target cells are labelled with mouse (BALB/c) monoclonal IgG1 antibody against human and chimpanzee CD3 and mouse monoclonal IgG1 antibody against human and chimpanzee CD45, together with magnetic beads, leaving the surface of the isolated T cells free of antibody binding. The procedure is performed at room temperature. Downstream applications include flow cytometry analysis, cell culture, and DNA/RNA purification, and the cells are intended for use in complement-dependent cytotoxicity (CDC) and flow cytometric crossmatch (FCXM) assays in HLA laboratories.
