Supporting the cloning and expansion of human and mouse T lymphocytes carrying neomycin-resistance transgenes, this kit from STEMCELL Technologies combines the methylcellulose-based ClonaCell semi-solid medium with a G418 (neomycin) selection supplement and a matched expansion medium. The single-cell-to-clone workflow supports colony outgrowth under continuous drug selection, removing the need for irradiated feeder layers during clone derivation.

Researchers use the system to isolate antigen-specific CD4+ and CD8+ T-cell clones from bulk cultures, to validate TCR-transduced populations after retroviral or lentiviral delivery, and to prepare clonal material for downstream assays such as cytokine profiling, cytotoxicity testing, and tetramer binding studies. Combining semi-solid outgrowth with cytokine-driven expansion shortens the path from a single transfected T cell to a working clone ready for functional characterization.

Components are shipped refrigerated and should be stored refrigerated upon arrival; working stocks of cytokine supplements are typically aliquoted to limit freeze-thaw cycles.
