ClonaCell-HY for Hybridomas, Kit

Generate monoclonal antibody-producing hybridoma cell lines using a complete semi-solid methylcellulose-based selection workflow. The STEMCELL Technologies ClonaCell-HY kit provides a full system for selecting and cloning hybridomas following PEG-mediated fusion of mouse myeloma cells with splenocytes. Three ready-to-use media are supplied: Medium A for pre-fusion cell preparation and plating, Medium B containing HAT selection agents suspended in methylcellulose to allow colony growth from successfully fused hybridomas while suppressing unfused myeloma and B-lymphocyte survival, and Medium C for liquid expansion of picked clones before downstream screening. Individual antibody-secreting colonies appear as discrete clusters embedded in the semi-solid matrix and can be lifted with a pipette or cloning ring for transfer to ELISA or flow cytometry assays. The semi-solid format supports single-step cloning in 6-well or 24-well plates and reduces the hands-on labor typically associated with limiting dilution methods. Store all components at -20 degrees C; thaw Medium B overnight at 4 degrees C before use and avoid repeated freeze-thaw cycles to preserve selection activity.
