Anti-Rat Nestin, Rat401, 100µg

A mouse monoclonal IgG1 antibody (clone Rat401) from STEMCELL Technologies targets nestin and reacts with both mouse and rat species. The unconjugated antibody was raised against nestin purified from embryonic rat spinal cord and is purified by affinity chromatography. It is supplied as a clear liquid in a phosphate-buffered solution at pH 7.2 containing 0.09% sodium azide.

Nestin is a > 170 kDa Class VI intermediate filament protein expressed in neuroepithelial stem and progenitor cells, as well as in other cell types including pancreatic islet progenitor cells, angiogenic endothelial cells, glioma cells, and bone marrow mesenchymal stem cells. The protein is required for the survival, renewal, and proliferation of neural progenitor cells and may also play a role in growth cone guidance during neuronal differentiation. Expression of nestin is down-regulated following neural differentiation, and within the adult nervous system it is limited mainly to progenitor cells in the cortical subventricular zone, the hippocampal dentate gyrus, dorsal root ganglia satellite cells, and a subpopulation of Schwann cells.

Immunocytochemistry (ICC) is a verified application, while ICC, immunofluorescence (IF), immunohistochemistry (IHC), and Western blotting (WB) are reported in the literature. Suggested starting concentrations are ≤ 10 µg/mL for ICC/IF and ≤ 2 µg/mL for WB, with titration recommended for optimal performance. The antibody is verified for labeling neural stem and progenitor cells cultured with the NeuroCult™ Proliferation Kit (Mouse & Rat) and the NeuroCult™ SM1 Neuronal Culture Kit.
