Description

This biotin-conjugated monoclonal antibody from STEMCELL Technologies, clone GL3, is an Armenian Hamster IgG2, kappa antibody that reacts with the δ chain of the murine T cell receptor γ/δ (TCRγ/δ). TCRγ/δ is a subtype of the T cell receptor involved in recognition of both peptide and lipid antigens. The GL3 antibody recognizes an epitope in the constant region of the δ chain and can reportedly activate TCRγ/δ+ cells. The antibody is reactive with mouse (Gene ID: 110066/110067) and was generated using intra-epithelial lymphocytes from C57BL/6J mice as the immunogen.

TCRγ/δ comprises a heterodimer (~ 80 kDa in humans) of disulfide-linked γ and δ subunits that associates with CD3 on the cell surface and is a member of the immunoglobulin superfamily. It is expressed on a subpopulation of T cells in the circulation but may be found on up to 50% of T cells in epithelial cell-rich tissues, where these cells contribute to oral and tumor-associated tolerance as well as autoimmune disease, and serve as a link between adaptive and innate immune responses. Most γ/δ T cells are CD4-/CD8-, though some express CD8.

The antibody was purified by affinity chromatography and conjugated with biotin under optimal conditions; the solution is free of unconjugated biotin. It is supplied at 0.5 mg/mL in an aqueous buffer containing 0.09% sodium azide and may contain carrier protein/stabilizer. Verified applications include cell separation (CellSep) and flow cytometry (FC); reported applications also include immunofluorescence (IF) and immunohistochemistry (IHC). For flow cytometry, the suggested use is ≤ 0.25 μg per 1 x 10^6 cells in 100 μL volume, with titration recommended for optimal performance. The product is stable at 2 - 8°C when stored undiluted; do not freeze.
