Description

This biotin-conjugated monoclonal antibody from STEMCELL Technologies, clone GL3, is an Armenian hamster IgG2 (kappa) reagent raised against the murine T cell receptor γ/δ (TCR gamma/delta). The clone reacts with an epitope in the constant region of the δ chain and is reported to activate TCRγ/δ+ cells. TCRγ/δ forms a disulfide-linked γ/δ heterodimer that associates with CD3 on the cell surface and is a member of the immunoglobulin superfamily, recognising both peptide and lipid antigens.

γ/δ T cells are found in the thymus, epidermis, intestinal and pulmonary epithelia, peritoneum, peripheral lymphoid tissues and reproductive organ mucosa. They account for a subpopulation of circulating T cells but may represent up to 50% of T cells in epithelial-rich tissues. Most γ/δ T cells are CD4-/CD8-, though some express CD8, and the dendritic epidermal T-cell subset is CD90+ (Thy-1+). The antibody was generated using intra-epithelial lymphocytes from C57BL/6J mice as immunogen and has been verified for cell separation and flow cytometry, with reported use in immunofluorescence and immunohistochemistry. It has also been validated for purity assessments of cells isolated with EasySep kits, including the EasySep Mouse T Cell Isolation Kit.

The reagent is purified by affinity chromatography and supplied in an aqueous buffer containing 0.09% sodium azide, free of unconjugated biotin. The suggested use for flow cytometry is ≤ 0.25 μg per 1 × 10⁶ cells in 100 μL; titration is recommended for optimal performance. The product is stable at 2 - 8°C when stored undiluted and should not be frozen.
