Anti-Ms SSEA-3, MC-631, AD, 25tests

STEMCELL Technologies offers a rat monoclonal IgM, kappa antibody (clone MC-631) conjugated to Alexa Fluor® 488 that recognizes the stage-specific embryonic antigen-3 (SSEA-3), specifically a galactosylgloboside epitope. The antibody was raised against four- to eight-cell stage mouse embryos and shows reactivity with human, mouse, rat, and rhesus samples. SSEA-3 is expressed on the surface of human embryonal carcinoma (EC), embryonic germ (EG), undifferentiated embryonic stem (ES), and induced pluripotent stem (iPS) cells, as well as rhesus monkey ES cell lines; no immunoreactivity is evident with undifferentiated mouse EC, EG, and ES cells. Expression is downregulated following differentiation of human ES and EC cells, whereas differentiation of mouse ES and EC cells may be accompanied by an increase in SSEA-3 expression.

Verified applications include flow cytometry (FC), immunocytochemistry (ICC), and immunofluorescence (IF), with immunohistochemistry (IHC) reported in published studies. The clone has been verified for labeling human ES and iPS cells maintained in TeSR™-E8™, mTeSR™1, and TeSR™2 media and for purity assessments of cells isolated with EasySep™ kits, including the TRA-1-60 Positive Selection Kit. The product is supplied in a phosphate-buffered solution at pH 7.2, containing 0.09% sodium azide and 0.2% (w/v) bovine serum albumin, and was purified by affinity chromatography prior to conjugation. It should be stored undiluted at 2–8 °C, protected from prolonged exposure to light, and not frozen. The suggested use is 5 μL per 1 × 10^6 cells in 100 μL volume (or per 100 μL of whole blood) for flow cytometry, and a 100× dilution for ICC/IF; titration is recommended for optimal performance in each application.
