Description

From STEMCELL Technologies, this unconjugated mouse monoclonal IgM, kappa antibody, clone MC-480, was raised against X-irradiated mouse F9 teratocarcinoma cells and recognizes the terminal carbohydrate epitope of stage-specific embryonic antigen-1 (SSEA-1), also referred to as CD15. The reagent reacts with human, mouse and rat SSEA-1 (Gene ID: 14345).

SSEA-1 is displayed on a high-molecular-mass (> 200 kDa) surface glycoprotein expressed by early mouse embryos, mouse embryonal carcinoma (EC) and embryonic stem (ES) cells, and mouse and human embryonic germ (EG) cells. It is absent from undifferentiated human EC, ES and induced pluripotent stem (iPS) cells and from rhesus monkey ES cell lines. On mouse ES cells, expression decreases upon differentiation, whereas on human cells it is upregulated during differentiation. The same carbohydrate epitope is also found on adult human granulocytes and monocytes (CD15) and has been linked to cell adhesion, migration and the regulation of cell differentiation.

Verified applications include cell separation (CellSep), flow cytometry, immunocytochemistry, immunofluorescence microscopy and Western blotting; additional reported uses include immunohistochemistry and immunoprecipitation. The antibody has been verified for labeling human ES and iPS cells cultured in TeSR™-E8™, mTeSR™1 and TeSR™2. It is provided in phosphate-buffered solution, pH 7.2, containing 0.09% sodium azide and was purified by affinity chromatography. The product remains stable when stored undiluted at 2 – 8 °C; it should not be frozen.
