Anti-Ms NK1.1, PK136, PS, 25µg

A mouse monoclonal IgG2a, kappa antibody from STEMCELL Technologies, clone PK136, conjugated to PerCP-Cy5.5 and directed against murine NK1.1 (CD161). The target is an approximately 80 kDa homodimeric type 2 transmembrane glycoprotein and C-type lectin receptor encoded by the Klrb1b and Klrb1c genes, expressed on NK cells, NK-T cells, and rare subsets of T cells and monocytes in select strains of mice. CD161b is expressed only by Swiss NIH and SJL mice, while CD161c is expressed by strains such as C57BL, FVB/N and NZB, but not by A, AKR, BALB/c, CBA/J, C3H, C57BR, C58, DBA/1, DBA/2, NOD, SJL or 129. NK1.1 has functional roles in modulating NK cell activation and proliferation, induction of interferon-gamma production, and release of cytotoxic granules, and its cell-surface expression is specifically upregulated by IL-12.

The antibody was raised in BALB/c mice using NK-1+ cells from mouse spleen and bone marrow as the immunogen, purified by affinity chromatography, and supplied in phosphate-buffered saline (pH 7.2) containing 0.09% sodium azide and 0.1% gelatin, free of unconjugated PerCP-Cy5.5. It has been verified and reported for flow cytometry; for detection of NK cells in NK1.1- strains, the antibody clone DX5 is recommended as an alternative NK cell marker. For flow cytometry the suggested use is ≤ 0.25 μg per 1 x 10e6 cells in 100 μL volume, and titration for optimal performance is recommended.

STEMCELL Technologies states the product is stable at 2 - 8°C when stored undiluted; it should not be frozen and should be protected from prolonged exposure to light. The antibody clone has been verified for purity assessments of cells isolated from NK1.1+ mouse strains such as C57BL/6, FVB/N, NZB, Swiss NIH, and SJL with the company's EasySep kits.
