Description

A monoclonal antibody from STEMCELL Technologies, clone PK136, reacts with murine NK1.1, also known as CD161. NK1.1 is described as an approximately 80 kDa homodimeric type 2 transmembrane glycoprotein and C-type lectin receptor expressed on NK cells, NK-T cells, and rare subsets of T cells and monocytes in select strains of mice. The antigen is encoded by the Klrb1b and Klrb1c genes, which specify the CD161b and CD161c polypeptides, respectively. CD161b is expressed only by Swiss NIH and SJL mice, while CD161c is expressed by strains such as C57BL, FVB/N and NZB, but not by A, AKR, BALB/c, CBA/J, C3H, C57BR, C58, DBA/1, DBA/2, NOD, SJL or 129 mice.

NK1.1 has functional roles in modulating the activation and proliferation of NK cells, the induction of interferon-γ production, and the release of cytotoxic granules, with cell surface expression specifically upregulated by IL-12. The antibody is of mouse origin with an IgG2a, kappa isotype. It is offered in multiple formats including Alexa Fluor 488, APC, biotin, FITC, PE, PerCP-Cyanine5.5 and unconjugated. Cited applications include flow cytometry, cell separation, CyTOF, depletion, functional assays, immunocytochemistry, immunofluorescence, immunohistochemistry, and immunoprecipitation. For detection of NK cells in NK1.1-negative strains, the manufacturer recommends the antibody clone DX5, which recognises CD49b (integrin α2).
