Anti-Ms NK1.1, PK136, BT, 500µg

This biotin-conjugated monoclonal antibody from STEMCELL Technologies, raised in a BALB/c mouse host, is clone PK136 of the mouse IgG2a, kappa isotype. It was generated against NK-1+ cells from mouse spleen and bone marrow and recognises murine NK1.1 (CD161), an approximately 80 kDa homodimeric type 2 transmembrane glycoprotein and C-type lectin receptor. The target is expressed on NK cells, NK-T cells and rare subsets of T cells and monocytes in select mouse strains, and is encoded by the Klrb1b and Klrb1c genes specifying the CD161b and CD161c polypeptides. CD161b is restricted to Swiss NIH and SJL mice, while CD161c is found in C57BL, FVB/N and NZB strains but not in A, AKR, BALB/c, CBA/J, C3H, C57BR, C58, DBA/1, DBA/2, NOD, SJL or 129. NK1.1 contributes to NK cell activation and proliferation, interferon-γ induction, and cytotoxic granule release, and its surface expression is specifically upregulated by IL-12.

Verified applications include cell separation (CellSep) and flow cytometry (FC), with additional reported use in immunofluorescence microscopy (IF) and immunohistochemistry (IHC). The reagent has been verified for purity assessments of cells from NK1.1+ mouse strains including C57BL/6, FVB/N, NZB, Swiss NIH and SJL using EasySep™ kits. For flow cytometry, the suggested use is ≤ 0.25 μg per 1 x 10e6 cells in 100 μL, with titration recommended for each application. The antibody is supplied at 0.5 mg/mL in a phosphate-buffered solution at pH 7.2 containing 0.09% sodium azide, purified by affinity chromatography and conjugated with biotin under conditions that leave the solution free of unconjugated biotin.

For storage, the product should be kept undiluted at 2 - 8°C and must not be frozen. For detection of NK cells in NK1.1-negative strains, the manufacturer recommends clone DX5, which recognises CD49b (integrin α2).
