Description

From STEMCELL Technologies, this mouse monoclonal IgG2a, kappa antibody (Clone PK136) targets murine NK1.1 (CD161), an approximately 80 kDa homodimeric type 2 transmembrane glycoprotein and C-type lectin receptor encoded by the Klrb1b and Klrb1c genes (Gene ID 17059). The antibody is conjugated with PE (Phycoerythrin) and was generated in a (C3H x BALB/c) F1 hybrid mouse host using NK-1+ cells from mouse spleen and bone marrow as the immunogen.

NK1.1 is expressed on NK cells, NK-T cells, and rare subsets of T cells and monocytes in select mouse strains. The CD161b isoform is found only in Swiss NIH and SJL mice, while CD161c is expressed by strains such as C57BL, FVB/N, and NZB. The antigen has functional roles in modulating NK cell activation and proliferation, inducing interferon-γ production, and triggering cytotoxic granule release; its surface expression is specifically upregulated by IL-12.

Purified by affinity chromatography and formulated in phosphate-buffered saline (pH 7.2) containing 0.09% sodium azide and 0.1% gelatin, the solution is free of unconjugated PE. The antibody is verified for flow cytometry (FC), with a suggested use of ≤ 0.5 μg per 1 × 10^6 cells in 100 μL, and titration is recommended for optimal performance. For NK1.1-negative strains, Anti-Mouse CD49b Antibody, Clone DX5 is recommended as an alternative NK cell marker. The product is stable at 2–8 °C when stored undiluted; it should not be frozen and should be protected from prolonged exposure to light.
