Description

Offered by STEMCELL Technologies, this FITC-conjugated mouse monoclonal IgG2a antibody (clone PK136) recognises mouse NK1.1 (CD161), an approximately 80 kDa homodimeric type 2 transmembrane glycoprotein and C-type lectin receptor expressed on NK cells, NK-T cells, and rare subsets of T cells and monocytes in select mouse strains. The antigen is encoded by the Klrb1b and Klrb1c genes, which specify the CD161b and CD161c polypeptides; CD161c is found in strains such as C57BL, FVB/N, and NZB, while CD161b is restricted to Swiss NIH and SJL mice, and several common strains — including A, AKR, BALB/c, CBA/J, C3H, C57BR, C58, DBA/1, DBA/2, NOD, and 129 — do not express NK1.1.

NK1.1 contributes to NK cell activation and proliferation, induction of interferon-γ production, and release of cytotoxic granules, with surface expression specifically upregulated by IL-12. The antibody was raised in a (C3H x BALB/c) F1 hybrid mouse against NK-1+ cells from mouse spleen and bone marrow, and is of the IgG2a, kappa isotype. Flow cytometry is the verified application, while immunofluorescence microscopy and immunohistochemistry have been reported in published studies. For detection of NK cells in NK1.1-negative strains, the manufacturer suggests an alternative anti-mouse CD49b antibody (clone DX5).

The reagent is supplied at 0.5 mg/mL in phosphate-buffered saline (pH 7.2) containing 0.09% sodium azide and 0.1% gelatin, and has been purified by affinity chromatography with no unconjugated FITC remaining. For flow cytometry, the suggested use is ≤ 0.5 μg per 1 × 10^6 cells in 100 μL. The product should be stored undiluted at 2 - 8 °C, protected from prolonged exposure to light and not frozen, and remains stable until the expiry date on the label.
