Anti-Ms Ly-6G, 1A8, BT, 500µg

A rat monoclonal antibody from STEMCELL Technologies targets the mouse Ly-6G antigen, also referred to as Gr-1, GR1, or Ly6G. Clone 1A8 binds specifically to Ly-6G, a 21–25 kDa GPI-anchored protein that, together with the structurally related Ly-6C protein, comprises the granulocyte receptor-1 antigen (Gr-1). The antibody is of the IgG2a, kappa isotype and was raised in the Rat (LEW) strain against a mouse Ly-6G-transfected EL-4J cell line. In contrast to clone RB6-8C5, which reacts with both Ly-6G and Ly-6C, clone 1A8 has been reported to detect Ly-6G-expressing granulocytes in peripheral blood without also binding Ly-6C-expressing lymphocytes, monocytes, and dendritic cells. Gr-1 is expressed on monocytes, neutrophils, and subsets of macrophages, plasmacytoid dendritic cells, and T cells, and is described as a useful marker for neutrophils in the periphery.

This biotin-conjugated reagent has been verified for flow cytometry, and representative data show staining of C57BL/6 mouse bone marrow cells labeled with the biotin conjugate followed by streptavidin APC. The antibody has been verified for purity assessments of cells isolated with EasySep kits, including the EasySep Mouse Neutrophil Enrichment Kit, supporting its use in the immunomagnetic negative isolation of untouched mouse neutrophils. For flow cytometry, the suggested use is ≤ 0.25 μg per 1 × 10^6 cells in 100 μL. The product is supplied as a phosphate-buffered solution, pH 7.2, containing 0.09% sodium azide, and has been purified by affinity chromatography and conjugated with biotin under optimal conditions, with the solution free of unconjugated biotin. It is stable at 2–8°C when stored undiluted and should not be frozen.
