Description

A rat monoclonal IgG1, kappa antibody from STEMCELL Technologies, clone XMG1.2, conjugated to PE (phycoerythrin) and specific for mouse interferon-gamma (IFN-γ). IFN-γ is a glycosylated type II interferon that exists as a homodimer of ~20 kDa subunits in its active form, produced primarily by activated T lymphocytes and natural killer (NK) cells in response to antigens including alloantigens, mitogens, and enterotoxins. The cytokine binds with high affinity to the α subunit of the IFN-γ receptor, leading to ligand-induced assembly of a tetrameric α2β2 receptor complex that activates the JAK-STAT pathway, with the receptor expressed on the surface of all nucleated cells.

The XMG1.2 antibody is suitable for detection of intracellular IFN-γ by flow cytometry when paired with an appropriate secondary antibody, and is widely used in various assays to neutralize the functional activity of IFN-γ. Verified and reported applications are both flow cytometry (FC). The antibody was generated against mouse recombinant IFN-γ expressed in E. coli and purified by affinity chromatography prior to conjugation with PE; the solution is free of unconjugated PE.

The reagent is supplied in phosphate-buffered saline, pH 7.2, containing 0.09% sodium azide and 0.1% gelatin, and remains stable at 2 - 8°C when stored undiluted. It should not be frozen and should be protected from prolonged exposure to light.
