Description

Rat monoclonal antibody from STEMCELL Technologies, clone MEL-14, reacts with mouse CD62L (also known as L-Selectin or LECAM-1), a type I transmembrane glycoprotein of approximately 74–95 kDa expressed on the surface of most leukocytes. The antibody is available in unconjugated form as well as conjugated to Alexa Fluor® 488, APC, biotin, FITC, PE, and PerCP-Cy5.5. It is of the IgG2a, kappa isotype and is raised in rat.

CD62L mediates leukocyte–endothelial cell interactions through association with ligands including CD34, glyCAM-1, MAdCAM-1, and PSGL-1, supporting lymphocyte rolling on the vascular endothelium, trafficking of lymphocytes to lymph nodes, and homing of lymphocytes and neutrophils to sites of inflammation. Murine CD62L is expressed on most thymocytes, with the highest levels on subsets of immunocompetent or dividing progenitor cells, as well as on neutrophils, eosinophils, monocytes, and subsets of B, T, and NK cells. The MEL-14 antibody can be used alongside other surface-marker antibodies to distinguish naïve, memory, and effector T cells, and has been shown to block lymphocyte migration to lymph nodes and to inhibit leukocyte rolling.

According to the manufacturer, the antibody has been verified for purity assessment of neutrophils and monocytes and is suited to flow cytometry workflows, including the immunomagnetic negative isolation of untouched mouse naïve CD4+ T cells, pan-naïve T cells, and naïve CD8+ T cells. Listed research applications include flow cytometry, functional assays, immunocytochemistry, immunofluorescence, immunohistochemistry, immunoprecipitation, and CyTOF.
