Description

STEMCELL Technologies offers this PE (Phycoerythrin)-conjugated rat monoclonal antibody (Clone GoH3) specific for mouse CD49f, also known as integrin α6. CD49f is an ~150 kDa transmembrane glycoprotein that associates non-covalently with CD29 (integrin β1) or CD104 (integrin β4) to form the heterodimeric receptors VLA-6 and α6β4, which bind the extracellular matrix protein laminin. The antibody is a disulfide-linked dimer comprising an ~120 kDa heavy chain and an ~30 kDa membrane-bound light chain. The reagent was raised in a Rat (SD) host and is of the IgG2a, kappa isotype, using mouse (BALB/c) mammary tumor cells as immunogen.

The antibody shows broad species reactivity, recognizing CD49f in human, mouse, rhesus, cynomolgus, baboon, chimpanzee, capuchin monkey, cat, cow, dog, horse, pig, rabbit, and sheep. Flow cytometry (FC) is verified, with FACS and FC further reported in the literature. The GoH3 clone reacts with an extracellular epitope on CD49f and reportedly blocks integrin α6 function in vivo and binding of integrin α6 to laminin in vitro. For flow cytometry, the suggested use is 5 μL per 1 x 10^6 cells in 100 μL or per 100 μL of whole blood, and titration is recommended for optimal performance in each application.

The antibody is supplied in a phosphate-buffered solution, pH 7.2, containing 0.09% sodium azide and 0.2% (w/v) bovine serum albumin, and was purified by affinity chromatography before conjugation with PE under optimal conditions; the solution is free of unconjugated PE and unconjugated antibody. The product is stable at 2 - 8°C when stored undiluted; it should not be frozen and should be protected from prolonged exposure to light.
