Anti-Ms CD49f, GoH3, PB, 25tests

This Pacific Blue™-conjugated monoclonal antibody from STEMCELL Technologies, clone GoH3, is raised in the rat (SD) and is of the IgG2a, kappa isotype. It targets CD49f (integrin α6), an approximately 150 kDa transmembrane glycoprotein that exists as a disulfide-linked dimer comprising an approximately 120 kDa heavy chain and an approximately 30 kDa membrane-bound light chain. CD49f pairs non-covalently with CD29 (integrin β1) or CD104 (integrin β4) to form the heterodimeric receptors VLA-6 and α6β4, which bind the extracellular matrix protein laminin. The clone was generated using mouse (BALB/c) mammary tumor cells as the immunogen and recognizes an extracellular epitope on CD49f; it has been reported to block integrin α6 function in vivo and to inhibit binding of integrin α6 to laminin in vitro.

CD49f is expressed on T cells, monocytes, platelets, placental trophoblasts, epithelial cells, and endothelial cells, and is considered the most important marker for selecting mouse mammary stem and progenitor cells. The antibody has been verified for use in flow cytometry, with a suggested use of 5 μL per 1 x 10^6 cells in 100 μL or per 100 μL of whole blood; titration is recommended for optimal performance. Reactivity is established with human, mouse, rhesus, cynomolgus, baboon, capuchin monkey, chimpanzee, cat, cow, dog, horse, pig, rabbit, and sheep samples.

The reagent is supplied as a phosphate-buffered solution at pH 7.2 containing 0.09% sodium azide and 0.2% (w/v) bovine serum albumin, and has been purified by affinity chromatography and conjugated under optimal conditions; the solution is free of unconjugated Pacific Blue™. For stability, the product should be stored undiluted at 2 - 8 °C, protected from prolonged light exposure and not frozen.
