Description
This rat monoclonal IgG2a, kappa antibody, supplied by STEMCELL Technologies, targets CD49f (integrin α6), an ~150 kDa transmembrane glycoprotein that associates non-covalently with CD29 (integrin β1) or CD104 (integrin β4) to form the heterodimeric receptors VLA-6 and α6β4, which bind the extracellular matrix protein laminin. The antibody, clone GoH3, was raised in Rat (SD) against mouse (BALB/c) mammary tumor cells and recognizes an extracellular epitope on CD49f, reportedly blocking integrin α6 function in vivo and binding of integrin α6 to laminin in vitro. CD49f is a disulfide-linked dimer of an ~120 kDa heavy chain and an ~30 kDa membrane-bound light chain, expressed on the surface of T cells, monocytes, platelets, placental trophoblasts, and epithelial and endothelial cells, and is considered the most important marker for selecting mouse mammary stem and progenitor cells.
The reagent is conjugated with Alexa Fluor® 488 and verified for flow cytometry (FC), with reported use in immunofluorescence (IF) and immunohistochemistry (IHC). It has been verified for labeling hematopoietic cells cultured in StemSpan™ SFEM and StemSpan™ H3000. Reactivity has been demonstrated in human, mouse, rhesus, cynomolgus, baboon, chimpanzee, capuchin monkey, cat, cow, dog, horse, pig, rabbit, and sheep samples. For flow cytometry, the suggested use is 5 μL per 1 x 10⁶ cells in 100 μL volume, or per 100 μL of whole blood, with titration recommended for optimal performance in each application.
The antibody is formulated in a phosphate-buffered solution at pH 7.2 containing 0.09% sodium azide and 0.2% (w/v) bovine serum albumin, purified by affinity chromatography, and supplied free of unconjugated Alexa Fluor® 488. It is stable at 2 - 8 °C when stored undiluted; it should not be frozen and should be protected from prolonged exposure to light. According to the manufacturer's safety data sheet, the clear liquid preparation is not classified as hazardous under the GHS or the 2012 OSHA Hazard Communication Standard, and is intended for use as a laboratory reagent.
