Description

This rat monoclonal antibody from STEMCELL Technologies, clone RA3-6B2, is supplied conjugated to FITC and recognises CD45R (B220), also referred to as protein tyrosine phosphatase receptor type C (PTPRC). The reagent reacts with human, mouse, and cat CD45R and is provided in a phosphate-buffered solution at pH 7.2 containing 0.09% sodium azide. The antibody was purified by affinity chromatography prior to conjugation, and the solution is free of unconjugated FITC.

The RA3-6B2 clone binds an epitope on the extracellular domain of the exon A-restricted isoform of mouse CD45, the largest CD45 isoform with a molecular mass in the 200–220 kDa range. CD45R is expressed throughout the B cell lineage, from early pro-B cells through to mature B cells, and its expression is downregulated during differentiation to plasma cells; it is also present on subsets of T cells, NK cells, dendritic cells, and lymphokine-activated killer (LAK) cells. The immunogen used to generate the antibody was Abelson murine leukemia virus-induced pre-B tumour cells, and the antibody is of the IgG2a, kappa isotype.

The reagent is verified for flow cytometry and has been reported for use in FACS, flow cytometry, immunofluorescence microscopy, and immunohistochemistry. It has been validated for purity assessments of cells isolated with EasySep kits, including the EasySep Mouse B Cell Isolation Kit. For flow cytometry the suggested use is ≤ 1 µg per 1 × 10^6 cells in a 100 µL volume, and titration is recommended for optimal performance. The product should be stored undiluted at 2–8 °C and protected from prolonged exposure to light; it must not be frozen. It is intended for research use only and is not for human or animal diagnostic or therapeutic applications.
