Description

A biotin-conjugated rat monoclonal antibody manufactured by STEMCELL Technologies, clone RA3-6B2, targets CD45R (also known as B220). The antibody reacts with an epitope on the extracellular domain of the exon A–restricted isoform of mouse CD45. CD45 is a member of the protein tyrosine phosphatase family, and its large cytoplasmic portion contains two tyrosine phosphatase domains (one enzymatically active) that modulate Src kinases such as Lck and Fyn. Alternative splicing of three extracellular exons (RA, RB and RC) gives rise to up to eight isoforms with molecular masses in the 200–220 kDa range; the exon A–restricted isoform, termed CD45R, contains the RA, RB, and RC regions and is the largest of these isoforms.

CD45R is expressed by the B cell lineage from early pro-B through mature B cells, with expression downregulated during differentiation to plasma cells, and is also found on subsets of T, NK, dendritic, and lymphokine-activated killer (LAK) cells. The antibody is of the IgG2a, kappa isotype and reacts with cat, human, and mouse samples. It is suited to applications including CyTOF, flow cytometry, functional assays, immunocytochemistry, immunofluorescence, immunohistochemistry, immunoprecipitation, and Western blotting, and is intended for research in immunology and transplantation.

The biotin (BT) format is suitable for large-scale flow cytometry panels and immunostaining workflows. The manufacturer recommends storage at 2–8°C, protection from light for fluorescent conjugates, and avoidance of repeated freeze–thaw cycles. The clone has been verified for purity assessments of cells isolated with EasySep™ kits, including the EasySep™ Mouse B Cell Enrichment Kit and EasySep™ Mouse B Cell Isolation Kit. For research use only.
