Anti-Ms CD45, 30-F11, UC, 500µg

A monoclonal antibody from STEMCELL Technologies, clone 30-F11, is directed against mouse CD45, a pan-leukocyte marker and type I transmembrane glycoprotein expressed on the surface of most hematopoietic cells, with the notable exceptions of mature erythrocytes, platelets and plasma cells [S1][S3]. The antibody reacts with an extracellular epitope found on all isoforms and both alloantigens (CD45.1 and CD45.2) of mouse CD45 [S1][S3]. CD45 is a member of the protein tyrosine phosphatase family and is involved in immunoregulatory processes including cell activation, growth, differentiation and oncogenic transformation; its large cytoplasmic portion contains two tyrosine phosphatase domains, one of which is enzymatically active, that modulate substrates such as the Src kinases Lck and Fyn [S3]. Alternative splicing of three exons (4, 5 and 6) encoding the extracellular RA, RB and RC polypeptide sequences gives rise to up to 8 isoforms with molecular masses in the range of 180–240 kDa [S3].

This unconjugated purified antibody (UC form) is supplied for labeling, conjugation or functional assays and is suitable for flow cytometry after secondary labeling, immunoprecipitation and Western blotting [S2]. Additional reported applications include CyTOF, immunocytochemistry, immunofluorescence, immunohistochemistry and functional assays [S3]. The host species is rat and the isotype is IgG2b, kappa [S3]. Conjugated formats offered for this clone include Alexa Fluor® 488, APC, Biotin, FITC, PE and PerCP-Cy5.5 [S3]. Recommended storage is 2–8°C [S2]. Flow cytometry validation data on C57BL/6 mouse splenocytes have been generated for the unconjugated, Alexa Fluor® 488, APC, Biotin, FITC, PE and PerCP-Cy5.5 conjugates, with a Rat IgG2b, kappa Isotype Control Antibody (Clone RTK4530) used as the negative control [S1].
