Description

This primary antibody from STEMCELL Technologies, Clone 30-F11, is a rat monoclonal IgG2b, kappa reagent (host species: rat; gene ID: 19264) that targets mouse CD45. It reacts with an extracellular epitope found on all isoforms and both alloantigens (CD45.1 and CD45.2) of CD45, a type I transmembrane glycoprotein expressed on the surface of most hematopoietic cells except mature erythrocytes, platelets, and plasma cells. CD45 expression is lost during differentiation of these cell types.

CD45 is a member of the protein tyrosine phosphatase family with roles in immunoregulatory processes including cell activation, growth, differentiation, and oncogenic transformation. Its large cytoplasmic region contains two tyrosine phosphatase domains, one enzymatically active, that modulate substrates such as the Src kinases Lck and Fyn. Alternative splicing of three exons (4, 5, 6) encoding the RA, RB, and RC extracellular polypeptide sequences gives rise to up to 8 isoforms with molecular masses of 180 to 240 kDa. The antibody is intended for immunology research and is validated for flow cytometry, CyTOF, functional assays, immunocytochemistry, immunofluorescence, immunohistochemistry, immunoprecipitation, and Western blot, with reactivity against granulocytes, hematopoietic stem and progenitor cells, lymphocytes, and myeloid cells. The reagent is available in multiple conjugations, including Alexa Fluor 488, APC, Biotin, FITC, PE, PerCP-Cyanine5.5, and unconjugated formats, and forms part of a workflow for immunomagnetic positive selection of mouse CD19+ cells.
