Anti-Ms CD45, 30-F11, PE, 50µg

This PE-conjugated monoclonal antibody, supplied by STEMCELL Technologies, is the clone 30-F11 raised in rats (LOU strain) against mouse thymus or spleen. It is of the IgG2b, kappa isotype and has been purified by affinity chromatography before conjugation with PE; the final solution is free of unconjugated PE and unconjugated antibody. The antibody reacts with an extracellular epitope found on all isoforms and both alloantigens (CD45.1 and CD45.2) of mouse CD45, a type I transmembrane glycoprotein expressed on the surface of most hematopoietic cells except mature erythrocytes, platelets, and plasma cells.

CD45 is a member of the protein tyrosine phosphatase family and is involved in immunoregulatory processes including cell activation, growth, differentiation, and oncogenic transformation. Its large cytoplasmic portion contains two tyrosine phosphatase domains, one of which is enzymatically active, that modulate intracellular substrates such as the Src kinases Lck and Fyn. Several isoforms arise from alternative splicing of exons 4, 5, and 6 (encoding the RA, RB, and RC polypeptide sequences), giving rise to up to 8 isoforms with molecular masses in the 180–240 kDa range. The reagent has been verified for flow cytometry and has also been verified for purity assessments of cells isolated with EasySep™ kits, including the EasySep™ Mouse CD19 Positive Selection Kit II.

Provided in a phosphate-buffered solution at pH 7.2 containing 0.09% sodium azide, the product should be stored undiluted at 2–8°C; it should not be frozen and should be protected from prolonged exposure to light. For flow cytometry, STEMCELL Technologies suggests using ≤ 0.25 μg per 1 × 10^6 cells in a 100 μL volume, with titration recommended for optimal performance for each application. According to the manufacturer's safety data sheet, the product is not classified as hazardous under the 2012 OSHA Hazard Communication Standard and presents as a clear liquid.
