Description

This biotin-conjugated mouse monoclonal IgG2a, kappa antibody from STEMCELL Technologies, clone A20, reacts with the mouse alloantigen CD45.1, also known as Ly5.1 and leukocyte common antigen. The A20 clone does not react with leukocytes expressing the CD45.2 alloantigen. CD45.1 is expressed by all hematopoietic cells except mature erythrocytes and platelets in mouse strains RIII, SJL/J, STS/A, and DA. Target alternative names include B220, CD45 antigen, CD45R, GP180, LCA, Ly5.1, Lyt4, Ptprc, RT7.1, and T200 (Gene ID 19264).

CD45 is a 180 - 240 kD type I transmembrane glycoprotein and a member of the protein tyrosine phosphatase (PTP) family, with two catalytic domains in the intracellular (COOH) region that regulate cellular signaling. The A20 antibody has been used extensively in adoptive cell transfer and bone marrow transplantation studies in mice to differentiate CD45.2- and CD45.1-expressing cells. It was raised against SJL mouse thymocytes and splenocytes and purified by affinity chromatography before conjugation with biotin, and the final solution is free of unconjugated biotin.

The reagent is supplied in a phosphate-buffered solution at pH 7.2 containing 0.09% sodium azide and should be stored at 2 - 8°C without freezing. Flow cytometry is the verified application, with FC, IHC, and IF reported in the literature. A suggested starting point for flow cytometry is ≤ 0.25 μg per 1 x 10^6 cells in a 100 μL volume, with titration recommended for optimal performance in each application.
