Description

From STEMCELL Technologies, the Anti-Mouse CD4 Antibody, Clone RM4-5 is a rat monoclonal IgG2a, kappa antibody supplied in a biotin-conjugated format. It reacts with mouse CD4, an ~55 kDa single-chain type 1 transmembrane glycoprotein belonging to the immunoglobulin (Ig) superfamily with four extracellular Ig-like domains. CD4 is expressed at relatively high levels by most thymocytes and a subpopulation of T cells (T-helper/inducer cells), and at lower levels on dendritic cells; in the mouse, CD4 is not expressed by monocytes/macrophages.

The antibody binds to CD4 and inhibits ligand binding in vitro, with binding reported to be blocked by the clone GK1.5 antibody. CD4 engages a non-polymorphic region of MHC II and acts as a co-receptor to the T cell receptor (TCR) in MHC II-restricted antigen recognition, enhancing the avidity of the TCR–MHC II–antigen complex association, and amplifies TCR signals through its intracellular domain's interaction with cytoplasmic tyrosine kinases such as Lck.

Applications include flow cytometry, immunofluorescence, and immunohistochemistry, as well as CyTOF, cell separation, functional assays, immunocytochemistry, immunoprecipitation, and Western blot. Available conjugations across the product family include Alexa Fluor 488, APC, biotin, FITC, PE, PerCP-Cyanine5.5, and unconjugated. This clone has been verified for purity assessments of cells isolated with EasySep™ kits and supports research areas including immunomagnetic positive selection of mouse CD25+ Tregs and immunomagnetic negative isolation of untouched mouse CD4+ T cells.
