Rat monoclonal IgG1, lambda antibody against mouse CD25 from STEMCELL Technologies, Clone PC61.5, conjugated to PerCP-Cy5.5 (Peridinin chlorophyll protein complex-Cyanine5.5). The clone was raised against the IL-2-dependent cytolytic mouse T cell clone B6.1 and is verified for flow cytometry (FC).

CD25 (also known as IL-2 receptor subunit alpha, IL-2Ra, IL-2Ralpha, IL-2-RA, Ly-43, p55, Tac) is an ~55 kDa type 1 transmembrane glycoprotein expressed on T and B cell progenitors, activated (but not resting) T and B cells, regulatory T (Treg) cells, dendritic cells, and activated monocytes and macrophages. Co-expression of CD25 with CD4 and FOXP3 is considered a phenotypic signature for Treg cells. CD25 associates with CD122 (IL-2Rβ) and CD132 (IL-2Rγ) to form the high-affinity IL-2 receptor, supporting receptor clustering and signaling in lymphocyte activation and proliferation.

Binding of PC61.5 inhibits IL-2 binding to both low- and high-affinity receptors, while the epitope recognized is distinct from the IL-2 binding site and from those of the 7D4 and 3C7 antibodies. The antibody has been verified for purity assessments of cells isolated with EasySep™ kits, including the EasySep™ Mouse T Cell Isolation Kit. It is supplied in phosphate-buffered solution, pH 7.2, containing 0.09% sodium azide and 0.1% gelatin, and was purified by affinity chromatography and conjugated under optimal conditions, with the solution free of unconjugated PerCP-Cy5.5. For flow cytometry, the suggested use is ≤ 0.25 μg per 1 x 10^6 cells in 100 μL; titration is recommended for optimal performance. Store undiluted at 2 - 8°C; do not freeze and protect from prolonged exposure to light.
