Description

A PE-conjugated monoclonal antibody from STEMCELL Technologies provides a tool for identifying and studying mouse CD25 (IL-2 receptor subunit α; Gene ID 16184) in flow cytometry. Clone PC61.5 is a rat IgG1, lambda reagent generated against the IL-2-dependent cytolytic mouse T cell clone B6.1 and reacts with the ~55 kDa type I transmembrane glycoprotein CD25 expressed on T and B cell progenitors, activated (but not resting) T and B cells, regulatory T (Treg) cells, dendritic cells, and activated monocytes and macrophages. The co-expression of CD25 with CD4 and FOXP3 is regarded as a phenotypic signature for Treg cells. PC61.5 binds an epitope distinct from the IL-2 binding site and from the epitopes recognized by clones 7D4 and 3C7, and binding of the antibody inhibits IL-2 engagement of both low- and high-affinity receptors, which may suppress IL-2-dependent lymphocyte proliferation.

The antibody has been verified for flow cytometry and used in purity assessments of cells isolated with EasySep kits, including the EasySep Mouse T Cell Isolation Kit. It is supplied as a liquid in phosphate-buffered solution at pH 7.2 containing 0.09% sodium azide and 0.1% gelatin, purified by affinity chromatography and conjugated under conditions that leave the solution free of unconjugated PE. For flow cytometry, a suggested use of ≤ 1 μg per 1 × 10⁶ cells in 100 μL is provided, and titration is recommended for each application.

The product is stable at 2 - 8 °C when stored undiluted; it should not be frozen and should be protected from prolonged exposure to light.
