Description

STEMCELL Technologies provides this PE (Phycoerythrin)-conjugated laboratory reagent, a rat monoclonal IgG1 (lambda) antibody raised against mouse CD25 and supplied as clone PC61.5. The product is intended for research use and is described in the manufacturer's safety data sheet as a clear liquid that is not a hazardous substance or mixture under the Globally Harmonized System (GHS).

CD25, also known as IL-2 receptor subunit alpha (IL-2Ra), Ly-43, p55, or Tac, is described as an approximately 55 kDa type 1 transmembrane glycoprotein expressed on T and B cell progenitors, activated (but not resting) T and B cells, regulatory T (Treg) cells, dendritic cells, and activated monocytes and macrophages. Co-expression of CD25 with CD4 and FOXP3 is considered a phenotypic signature for Treg cells. The PC61.5 clone has been reported to inhibit IL-2 binding to both low- and high-affinity receptors, with its epitope being distinct from the IL-2 binding site and from epitopes recognised by clones 7D4 and 3C7.

The reagent has been verified for use in flow cytometry and has also been reported in the same application. It is formulated in a phosphate-buffered solution at pH 7.2 containing 0.09% sodium azide and 0.1% gelatin, and was purified by affinity chromatography prior to PE conjugation. For flow cytometry, the manufacturer suggests using ≤ 1 µg per 1 × 10^6 cells in 100 µL, with titration recommended for optimal performance in each application. The product should be stored at 2 – 8°C, protected from prolonged exposure to light, and not frozen. STEMCELL Technologies states that the product is for research use only and is not intended for human or animal diagnostic or therapeutic uses unless otherwise stated.
