Description

This biotin-conjugated rat monoclonal IgG1, lambda antibody from STEMCELL Technologies targets mouse CD25, also known as the low-affinity interleukin-2 receptor α chain (IL-2Rα) and referred to by the alternative names Ly-43, p55, and Tac. The antibody, clone PC61.5, was raised against the IL-2-dependent cytolytic mouse T cell clone B6.1 and is supplied in a phosphate-buffered solution at pH 7.2 containing 0.09% sodium azide, purified by affinity chromatography and conjugated with biotin under optimal conditions so that the solution is free of unconjugated biotin.

CD25 is an approximately 55 kDa type I transmembrane glycoprotein expressed on T and B cell progenitors, activated (but not resting) T and B cells, regulatory T (Treg) cells, dendritic cells, and activated monocytes and macrophages, and its co-expression with CD4 and FOXP3 is considered a phenotypic signature for Treg cells. The epitope recognised by clone PC61.5 is distinct from the IL-2 binding site and from the epitopes recognised by antibodies 7D4 and 3C7. Binding of the antibody has been reported to inhibit binding of IL-2 to low- and high-affinity receptors.

Verified applications include flow cytometry, with immunohistochemistry also reported in the literature. The antibody has been verified for purity assessments of cells isolated with EasySep kits, and the suggested use for flow cytometry is ≤ 0.25 μg per 1 × 10^6 cells in 100 μL, with titration recommended for optimal performance for each application. The product should be stored at 2–8°C undiluted and should not be frozen, and is intended for research use only.
