Description

From STEMCELL Technologies, this biotinylated anti-mouse CD25 antibody (clone PC61.5) is a rat IgG1, lambda reagent supplied for laboratory research use. It recognizes murine CD25, the low-affinity interleukin-2 receptor α chain (IL-2Rα), described as an ~55 kDa type 1 transmembrane glycoprotein.

CD25 is expressed on T and B cell progenitors, on activated (but not resting) T and B cells, on T regulatory (Treg) cells, on dendritic cells, and on activated monocytes and macrophages. Co-expression of CD25 with CD4 and FOXP3 is regarded as a phenotypic signature of Treg cells. Although CD25 on its own has low affinity for IL-2, it associates with CD122 (IL-2Rβ) and CD132 (IL-2Rγ) to assemble the high-affinity IL-2 receptor, and it is required for receptor clustering and the signaling that drives lymphocyte activation and proliferation. Binding of the PC61.5 antibody blocks IL-2 binding to both the low- and high-affinity receptors, which may inhibit IL-2-dependent proliferation. The PC61.5 epitope is distinct from the IL-2 binding site and from the epitopes recognized by the 7D4 and 3C7 antibodies.

The manufacturer's safety data sheet describes the product as a clear liquid that is not classified as a hazardous substance or mixture according to the Globally Harmonized System (GHS). Listed applications include flow cytometry, immunohistochemistry, immunoprecipitation, functional assays, cell depletion, and neutralization/blocking.
