Description

The PC61.5 clone is a rat monoclonal IgG1, lambda antibody that recognizes mouse CD25, the ~55 kDa low-affinity interleukin-2 receptor α chain (IL-2Rα). CD25 is a type 1 transmembrane glycoprotein expressed on T and B cell progenitors, activated (but not resting) T and B cells, regulatory T (Treg) cells, dendritic cells, and activated monocytes and macrophages. Co-expression of CD25 with CD4 and FOXP3 is considered a phenotypic signature for Treg cells, and CD25 associates with CD122 (IL-2Rβ) and CD132 (IL-2Rγ) to form the high-affinity IL-2R that supports lymphocyte activation and proliferation.

Binding of the PC61.5 antibody inhibits IL-2 interaction with both low- and high-affinity receptors, which can suppress IL-2-dependent proliferation, and the recognized epitope is distinct from the IL-2 binding site as well as from the epitopes recognized by the 7D4 and 3C7 clones. The antibody was raised against the IL-2-dependent cytolytic mouse T cell clone B6.1 and is supplied by STEMCELL Technologies as an unconjugated reagent purified by affinity chromatography in a phosphate-buffered solution at pH 7.2 containing 0.09% sodium azide.

The reagent has been verified for flow cytometry and is reported for use in blocking, depletion, functional assays, immunohistochemistry, and immunoprecipitation, and has also been verified for purity assessments of cells isolated with EasySep™ kits. It is stable at 2 - 8 °C when stored undiluted and should not be frozen.
