Anti-Ms CD25, 3C7, PE, 25µg

This PE-conjugated monoclonal antibody from STEMCELL Technologies, clone 3C7, targets murine CD25 (interleukin-2 receptor alpha chain; IL-2Ralpha), an approximately 55 kDa type 1 transmembrane glycoprotein expressed on the surface of T and B cell progenitors and on activated mature B and T cells. The reagent is a rat (LEW) monoclonal of the IgG2b, kappa isotype, raised against an IL-2-dependent BALB/c mouse helper T cell clone (HT-2). Its epitope is distinct from those recognized by the anti-CD25 antibody clones 7D4 and PC61.

CD25 is widely used as a marker to identify CD4+/FoxP3+ regulatory T cells in mice. On its own, CD25 has low affinity for its IL-2 ligand but forms dimers with CD122 (IL-2Rbeta) and CD132 (IL-2Rgamma) that together associate to form the IL-2R receptor, which binds IL-2 with high affinity. CD25 increases the specificity and affinity of IL-2 binding and is necessary for receptor clustering and signal transduction by the complex. Binding of the 3C7 antibody inhibits binding of IL-2 to free CD25 and to the IL-2R receptor. Verified applications include cell separation (CellSep) and flow cytometry (FC), and the clone has been validated for purity assessments of cells isolated with EasySep kits, including the EasySep Mouse T Cell Enrichment Kit.

The antibody was purified by affinity chromatography and conjugated with PE under optimal conditions, and the solution is free of unconjugated PE and unconjugated antibody. It is formulated in a phosphate-buffered solution at pH 7.2 containing 0.09% sodium azide. For flow cytometry, the suggested use is less than or equal to 1 ug per 1 x 10^6 cells in 100 uL volume, with titration recommended for optimal performance in each application. The product is stable at 2-8 degrees C when stored undiluted; it should not be frozen and should be protected from prolonged exposure to light. Alternative designations for the target antigen include IL-2Ralpha, Ly-43, p55, and Tac (Gene ID: 16184).
