Description

A PE-conjugated rat monoclonal antibody (clone 3C7; isotype IgG2b, kappa) from STEMCELL Technologies reacts with murine CD25, the ~55 kDa interleukin-2 receptor α chain (IL-2Rα; also known as p55, Tac, and Ly-43) expressed on T and B cell progenitors and on activated mature B and T cells. CD25 is widely used as a marker to identify CD4+/FoxP3+ regulatory T cells in mice. The antibody was raised in a Rat (LEW) host against the IL-2-dependent BALB/c mouse helper T cell clone HT-2, purified by affinity chromatography, and conjugated with PE under optimal conditions, with the solution free of unconjugated PE and unconjugated antibody.

Applications verified by the manufacturer include flow cytometry and cell separation, including purity assessments of cells isolated with EasySep kits. The 3C7 clone inhibits binding of IL-2 to free CD25 and to the IL-2R receptor, and its epitope is distinct from those recognized by the anti-CD25 clones 7D4 and PC61. The antibody is formulated in a phosphate-buffered solution at pH 7.2 containing 0.09% sodium azide and is supplied at 0.2 mg/mL. For flow cytometry, the suggested use is ≤ 1 µg per 1 x 10^6 cells in 100 µL. The product should be stored at 2–8°C, protected from prolonged exposure to light, and not frozen.
