Description

This FITC-conjugated monoclonal antibody from STEMCELL Technologies, clone 3C7, is a rat IgG2b, kappa reagent raised against mouse CD25 (also known as IL-2Rα, Ly-43, p55, and Tac). It reacts with murine CD25, an approximately 55 kDa type 1 transmembrane glycoprotein expressed on the surface of T and B cell progenitors and on activated mature B and T cells. CD25 has been used as a marker to identify CD4+/FoxP3+ regulatory T cells in mice, and mutations in CD25 result in severe immunodeficiency, underscoring its role in lymphocyte differentiation, activation, and proliferation, and in the maintenance of self-tolerance. The target forms dimers with CD122 (IL-2Rβ) and CD132 (IL-2Rγ) to create the high-affinity IL-2 receptor, and binding of the 3C7 antibody inhibits binding of IL-2 to free CD25 and to the IL-2R receptor. The recognized epitope is distinct from those bound by the anti-CD25 clones 7D4 and PC61.

The antibody was purified by affinity chromatography and conjugated with FITC; the solution is free of unconjugated FITC and is supplied in a phosphate-buffered solution at pH 7.2 containing 0.09% sodium azide. The immunogen was the IL-2-dependent BALB/c mouse helper T cell clone HT-2, and the host species is rat (LEW). Applications verified and reported include flow cytometry (FC), and the reagent has been verified for purity assessments of cells isolated with EasySep™ kits, including the EasySep™ Mouse T Cell Enrichment Kit. It is also used in immunomagnetic positive selection of mouse CD25+ regulatory T cells from splenocytes or other single-cell suspensions.

For flow cytometry, the suggested use is ≤ 1 µg per 1 × 10^6 cells in 100 µL, with titration recommended for optimal performance. The product is stable at 2–8 °C when stored undiluted; it should not be frozen and should be protected from prolonged exposure to light. For research use only.
