Description

A rat monoclonal antibody from STEMCELL Technologies, clone 2.4G2, targets a shared epitope on mouse Fc-gamma receptors CD16 (FcRIII) and CD32 (FcRII). The antibody is unconjugated and of the IgG2b, kappa isotype, generated using the mouse BALB/c macrophage J774 cell line as immunogen.

CD16 and CD32 serve as low-affinity receptors for the Fc portion of IgG, with CD32 displaying higher affinity for IgG aggregates and immune complexes. These receptors are expressed on B cells, monocytes, macrophages, NK cells, neutrophils, granulocytes, mast cells, and dendritic cells. Their engagement participates in phagocytosis, cytotoxicity, and modulation of immune responses. By blocking the interaction between IgG and CD16 or CD32, the 2.4G2 antibody is commonly applied as a pre-treatment to minimize non-specific binding of antibodies to FcR-bearing cells in flow cytometry, cell sorting, and immunofluorescence.

Flow cytometry is a verified application, with FA (Blocking), IF, and IP reported in the literature. The product is supplied in phosphate-buffered saline, pH 7.2, with low endotoxin (≤ 0.01 EU/μg as determined by the Limulus amebocyte lysate test) and no azide, supporting its use for in vivo studies in mice. It is purified by affinity chromatography and should be stored at 2 - 8°C without freezing. For flow cytometry, the suggested use is ≤ 1 μg per 1 x 10^6 cells in 100 μL volume, with titration recommended for optimal performance.
