Description

Rat monoclonal IgG2a, lambda antibody conjugated to R-phycoerythrin (PE) and offered by STEMCELL Technologies. Clone TC15-12F12.2 reacts with mouse CD150, also known as SLAM (signaling lymphocyte activation molecule) or IPO-3, an approximately 75 kDa type I transmembrane glycoprotein that functions as a cell adhesion molecule and/or co-receptor. It is differentially expressed by T cells, immature thymocytes, B cells, dendritic cells, macrophages, and endothelial cells, and its expression is rapidly induced upon activation of T cells, B cells, and dendritic cells. On T cells, synthesis is maintained on Th1 but not Th2 clones. CD150-mediated co-stimulation of TCR-activated T cells has been reported to enhance IFN-γ production by Th1 cells, a response that can be augmented by binding of the TC15-12F12.2 antibody, and the antigen is thought to mediate signal transduction by associating with the intracellular protein tyrosine phosphatase SHP-2.

The reagent was raised in rats of the LEW strain against a mouse SLAM-human IgG1 fusion protein, purified by affinity chromatography, and conjugated with PE under conditions that leave the solution free of unconjugated antibody and unconjugated PE. It is formulated in phosphate-buffered solution at pH 7.2 containing 0.09% sodium azide, and the manufacturer specifies storage at 2 - 8°C, with instructions not to freeze and to protect the product from prolonged exposure to light. CD150 is described as a useful marker, used in concert with other markers such as CD48 and CD41, for the prospective detection of multipotent hematopoietic stem cells, and it is not expressed on non-multipotent hematopoietic progenitor cells.

Flow cytometry (FC) is listed as a verified and reported application. For FC, the manufacturer suggests ≤ 0.25 μg per 1 x 10^6 cells in 100 μL, with titration recommended for optimal performance in each application. Representative data shown by the manufacturer demonstrate staining of C57BL/6 mouse splenocytes and bone marrow cells by flow cytometry, with parallel rat IgG2a, lambda PE isotype control comparisons. The associated gene identifier is 27218.
