Anti-Ms CD150, TC15-12F12.2, BT, 100µg

Rat monoclonal IgG2a antibody raised against mouse CD150 (signaling lymphocyte activation molecule, or SLAM), supplied by STEMCELL Technologies in a biotin-conjugated format (Clone TC15-12F12.2). CD150 is an ~75 kDa type I transmembrane glycoprotein that acts as a cell adhesion molecule and coreceptor in the immune response. According to the manufacturer, it is differentially expressed by T cells, immature thymocytes, B cells, dendritic cells, macrophages, and endothelial cells, with expression rapidly induced upon activation of T cells, B cells, and dendritic cells. Synthesis by T cells is maintained on Th1 but not Th2 clones, and CD150-mediated co-stimulation of TCR-activated T cells enhances IFN-γ production by Th1 cells, a response that can be augmented by binding of the TC15-12F12.2 antibody. CD150 is reported to mediate signal transduction by associating with the intracellular protein tyrosine phosphatase SHP-2, and it also functions in hematopoietic cell development as a useful marker for multipotent hematopoietic stem cells (in concert with other markers such as CD48 and CD41), while not being expressed on non-multipotent hematopoietic progenitor cells.

The antibody was raised in the rat (LEW) strain using a mouse SLAM-human IgG1 fusion protein as immunogen, then purified by affinity chromatography and conjugated with biotin under optimal conditions; the solution is free of unconjugated biotin. It is formulated in a phosphate-buffered solution at pH 7.2 containing 0.09% sodium azide. Flow cytometry is a verified application, with FC, IF, and IHC reported in the literature. For flow cytometry, the manufacturer suggests using ≤ 0.25 μg per 1 x 10^6 cells in 100 μL, and recommends titration for optimal performance for each application. The product is stable at 2 - 8°C when stored undiluted and should not be frozen.
