Description

STEMCELL Technologies offers this PE-conjugated rat monoclonal IgG2b, kappa antibody, raised by the M1/70 clone, which targets an ~170 kDa type 1 transmembrane glycoprotein. The antibody recognizes CD11b (also known as alphaM integrin, C3biR, CR3, Ly-40, Mac-1, or Mo1), which associates non-covalently with CD18 to form the heterodimeric Mac-1 receptor.

Reactivity has been demonstrated with human, mouse, rhesus, cynomolgus, baboon, chimpanzee, and rabbit samples. Through its interactions with ICAM-1 (CD54), ICAM-2 (CD102), ICAM-4 (CD242), iC3b, and fibrinogen, Mac-1 functions in processes including the adherence of neutrophils and monocytes to stimulated endothelium and phagocytosis of complement-coated particles. CD11b is expressed on the surface of granulocytes, monocytes, NK cells, dendritic cells, tissue macrophages, and subsets of T and B cells. The M1/70 antibody reportedly blocks iC3b binding to Mac-1.

Verified applications include cell separation (CellSep) and flow cytometry (FC), with reported use in FACS, immunocytochemistry, immunofluorescence microscopy, and immunohistochemistry. The immunogen used was C57BL/10 mouse splenocytes. The antibody is formulated in phosphate-buffered saline, pH 7.2, containing 0.09% sodium azide and 0.1% gelatin, and is supplied as a clear liquid at 0.2 mg/mL. For flow cytometry, the recommended use is ≤ 0.125 μg per 1 x 10^6 cells in 100 μL. Storage at 2 - 8°C is recommended; the product should not be frozen and should be protected from prolonged exposure to light.
