Anti-Ms CD11b, M1/70, PB, 25µg

STEMCELL Technologies offers a Pacific Blue™-conjugated rat monoclonal antibody (clone M1/70, IgG2b kappa) directed against CD11b. CD11b is an approximately 170 kDa type I transmembrane glycoprotein that associates non-covalently with CD18 to form the heterodimeric Mac-1 receptor. Mac-1 mediates adherence of neutrophils and monocytes to stimulated endothelium and phagocytosis of complement-coated particles through interactions with ICAM-1 (CD54), ICAM-2 (CD102), ICAM-4 (CD242), iC3b, and fibrinogen.

Flow cytometry is a verified application for this clone, with immunofluorescence additionally reported in the literature. The antibody clone has been verified for purity assessments of cells isolated with EasySep™ kits, including the EasySep™ Mouse Monocyte Isolation Kit. Reported species reactivity includes human, mouse, rhesus, cynomolgus, baboon, chimpanzee, and rabbit. CD11b is expressed on granulocytes, monocytes, NK cells, dendritic cells, tissue macrophages, and subsets of T and B cells, and has been used as a marker to distinguish naïve and memory CD8+ T cells. The M1/70 clone reportedly blocks iC3b binding to Mac-1.

The reagent is supplied as a phosphate-buffered solution at pH 7.2 containing 0.09% sodium azide and is purified by affinity chromatography, with the solution being free of unconjugated Pacific Blue™. For flow cytometry, a suggested use of ≤ 1 μg per 1 x 10^6 cells in 100 μL is recommended, with titration advised for optimal performance for each application.
