Anti-Hu TRA-1-60, TRA-1-60R, PE, 25tests

Manufactured by STEMCELL Technologies, this PE-conjugated mouse monoclonal antibody (clone TRA-1-60R, IgM, kappa) targets TRA-1-60 (podocalyxin), a >200 kDa pluripotent stem cell-specific surface protein expressed on undifferentiated human embryonic stem (ES), induced pluripotent stem (iPS), embryonal carcinoma (EC), and embryonic germ (EG) cells, as well as rhesus monkey ES cell lines. A soluble form of TRA-1-60 has been detected in serum of patients with embryonal carcinoma. The antibody was raised against the human embryonal carcinoma cell line 2102Ep cl.2A6 and shows reactivity against human, rhesus, and rabbit samples. The sialic acid-containing epitope is lost upon cell differentiation and is associated with podocalyxin, yet remains resistant to neuraminidase treatment. Suggested starting conditions are 5 μL per 1 x 10^6 cells in 100 μL for flow cytometry, or a 100X dilution for immunocytochemistry and immunofluorescence. Verified applications include cell separation, flow cytometry, immunocytochemistry, and immunofluorescence, and the reagent has been validated for labeling human ES and iPS cells cultured in TeSR-E8, mTeSR1, and TeSR2 media, as well as for purity assessments of cells isolated with EasySep kits.

The antibody is supplied as a clear liquid in a phosphate-buffered solution at pH 7.2 containing 0.09% sodium azide and 0.2% (w/v) bovine serum albumin, and has been purified by affinity chromatography and conjugated with PE under optimal conditions. It is stable at 2 - 8°C when stored undiluted; it should not be frozen and should be protected from prolonged exposure to light. According to the manufacturer's safety data sheet, the mixture is not classified as hazardous under the GHS or the 2012 OSHA Hazard Communication Standard and contains no substances considered hazardous at their given concentration.
