Description

The TRA-1-60R antibody from STEMCELL Technologies is a biotin-conjugated mouse monoclonal IgM, kappa antibody that recognizes TRA-1-60, a > 200 kDa pluripotent stem cell-specific protein. TRA-1-60 is expressed on the surface of undifferentiated human embryonic stem (ES) and induced pluripotent stem (iPS) cells, embryonal carcinoma (EC) cells, and embryonic germ (EG) cells, as well as rhesus monkey ES cell lines. A soluble form of TRA-1-60 has been detected in serum of patients with embryonal carcinoma.

The recognized epitope, which is lost upon cell differentiation, contains sialic acid and is associated with a large-molecular-mass transmembrane protein named podocalyxin (Gene ID: 5420). Although sialylated, the epitope recognized by TRA-1-60R is resistant to treatment with neuraminidase. The clone was raised against the human embryonal carcinoma cell line 2102Ep cl.2A6 and reacts with human, rhesus, and rabbit samples.

Flow cytometry (FC) is the verified application; reported applications include FC, immunocytochemistry (ICC), immunofluorescence microscopy (IF), immunohistochemistry (IHC), immunoprecipitation (IP), and Western blotting (WB). The clone has been verified for labeling human ES and iPS cells grown in TeSR™-E8™, mTeSR™1, and TeSR™2 media. The reagent is supplied at 0.5 mg/mL in a phosphate-buffered solution at pH 7.2 containing 0.09% sodium azide, free of unconjugated biotin. For flow cytometry, the suggested use is ≤ 0.25 µg per 1 x 10^6 cells in 100 µL. Store undiluted at 2 - 8°C; do not freeze. For research use only.
