Anti-Hu TRA-1-60, TRA-1-60R, AD, 100t

This mouse monoclonal IgM antibody from STEMCELL Technologies, Clone TRA-1-60R conjugated with Alexa Fluor® 488, targets TRA-1-60 (Podocalyxin), a > 200 kDa pluripotent stem cell-specific surface protein expressed on undifferentiated human embryonic stem (ES), induced pluripotent (iPS), embryonal carcinoma (EC), and embryonic germ (EG) cells, as well as rhesus monkey ES cell lines. The antibody is reactive with human, rhesus, and rabbit species.

The epitope recognized by the TRA-1-60R clone contains sialic acid associated with the podocalyxin transmembrane protein and, although sialylated, is resistant to neuraminidase treatment. The epitope is lost upon cell differentiation. The antibody was raised against the human embryonal carcinoma cell line 2102Ep cl.2A6 and purified by affinity chromatography before conjugation; the solution is free of unconjugated Alexa Fluor® 488.

Verified applications include flow cytometry (FC), immunocytochemistry (ICC), and immunofluorescence (IF). Suggested use is 5 μL per 1 x 10^6 cells in 100 μL for FC and a 100X dilution for ICC/IF. The reagent has been verified for labeling human ES and iPS cells cultured in TeSR™-E8™, mTeSR™1, and TeSR™2 media. The product is formulated as a phosphate-buffered solution at pH 7.2 containing 0.09% sodium azide and 0.2% (w/v) bovine serum albumin, appears as a clear liquid, and should be stored at 2 - 8°C undiluted; it should not be frozen and should be protected from prolonged light exposure.
