Description

A PE-conjugated monoclonal antibody from STEMCELL Technologies targets human OCT4 (OCT3), an ~40 kDa POU-family homeodomain transcription factor. The reagent is a mouse IgG2b, kappa antibody (Clone 3A2A20) raised against recombinant partial human OCT4 protein (amino acids 1–141), and is verified and reported for flow cytometry applications. According to the manufacturer, the antibody has been validated for labeling human ES and iPS cells grown in TeSR™-E8™ and mTeSR™1 culture systems.

OCT4 plays a key role in maintaining pluripotency by interacting with transcription factors such as SOX2 to regulate expression of downstream genes including FBX15, FGF-4, REX1, SOX2, and osteopontin. The marker is expressed in undifferentiated human embryonic stem (ES), induced pluripotent stem (iPS), embryonal carcinoma (EC), and embryonic germ (EG) cells, and is down-regulated during differentiation, making it a useful indicator of pluripotency as well as a marker for certain human malignant germ cell tumours. Expression of OCT4 together with other transcription factors has been used to reprogram somatic cells into iPS cells.

The product is supplied as a phosphate-buffered solution at pH 7.2, containing 0.09% sodium azide and 0.2% (w/v) bovine serum albumin. It was purified by affinity chromatography and conjugated with PE under optimal conditions, and the solution is free of unconjugated PE and unconjugated antibody. For flow cytometry, the suggested use is 5 μL per 1 x 10^6 cells in 100 μL, or per 100 μL of whole blood. The reagent is a clear liquid and is not classified as hazardous under GHS or the 2012 OSHA Hazard Communication Standard. STEMCELL Technologies recommends storage at 2–8°C when undiluted; the product should not be frozen and should be protected from prolonged exposure to light.
