Anti-Hu OCT4 (OCT3), 3A2A20, AD, 25tests

A mouse monoclonal IgG2b antibody from STEMCELL Technologies, conjugated to Alexa Fluor® 488, that recognizes human OCT4 (octamer-binding transcription factor 4; also known as OCT3 and OCT3/4), an ~40 kDa POU-family homeodomain transcription factor (Gene ID 5460). Clone 3A2A20 was raised against recombinant partial human OCT4 protein (amino acids 1 - 141). The antibody reacts with OCT4 expressed in undifferentiated human embryonic stem (ES), induced pluripotent stem (iPS), embryonal carcinoma (EC) and embryonic germ (EG) cells. OCT4 binds the octamer motif 5'- ATTTGCAT-3' and, through interactions with factors such as SOX2, helps maintain pluripotency by regulating expression of FBX15, FGF-4, REX1, SOX2 and osteopontin. Levels fall during differentiation, making this reagent a marker of pluripotency in stem cells and of certain human malignant germ cell tumours.

Flow cytometry is the verified application, with a suggested use of 5 μL per 1 x 10^6 cells in 100 μL volume, or per 100 μL of whole blood; titration is recommended for optimal performance. The clone has been verified for labeling human ES and iPS cells cultured in TeSR™-E8™, mTeSR™1 and TeSR™2. The product is supplied as a clear liquid in a phosphate-buffered solution at pH 7.2 containing 0.09% sodium azide and 0.2% (w/v) bovine serum albumin. According to the manufacturer's safety data sheet, the preparation is not classified as hazardous under the Globally Harmonized System (GHS) or the 2012 OSHA Hazard Communication Standard (29 CFR 1910.1200).

For storage, the reagent is stable at 2 - 8°C when kept undiluted; it should not be frozen and should be protected from prolonged exposure to light. STEMCELL Technologies Canada Inc.
