Anti-Hu OCT4 (OCT3), 3A2A20, AD, 100t

A mouse monoclonal IgG2b antibody from STEMCELL Technologies, clone 3A2A20, conjugated to Alexa Fluor® 488, targets human OCT4 (OCT3), an approximately 40 kDa homeodomain transcription factor of the POU family (alternative names include OCT-3, OCT-4, OCT3, octamer-binding transcription factor 4, POU domain class 5 transcription factor 1, POU5F1; Gene ID: 5460). The reagent was raised against recombinant partial human OCT4 protein corresponding to amino acids 1–141 and is reactive with human OCT4.

By binding the octamer motif 5'-ATTTGCAT-3', OCT4 helps maintain pluripotency in undifferentiated human embryonic stem (ES), induced pluripotent stem (iPS), embryonal carcinoma (EC), and embryonic germ (EG) cells, cooperating with factors such as SOX2 to regulate expression of genes including FBX15, FGF-4, REX1, SOX2, and osteopontin. OCT4 levels are down-regulated during differentiation, and it has emerged as a marker of pluripotency in stem cells as well as a marker for certain human malignant germ cell tumours. The antibody has been verified for flow cytometry (FC) labeling of human ES and iPS cells cultured in TeSR™-E8™ and mTeSR™1.

The product is supplied as a phosphate-buffered solution (pH 7.2) containing 0.09% sodium azide and 0.2% (w/v) bovine serum albumin, purified by affinity chromatography and conjugated with Alexa Fluor® 488 under optimal conditions, free of unconjugated dye. For flow cytometry, the suggested use is 5 μL per 1 × 10^6 cells in 100 μL, or per 100 μL of whole blood; titration is recommended for optimal performance in each application. The reagent is stable at 2 - 8°C when stored undiluted; it should not be frozen and should be protected from prolonged exposure to light. According to the manufacturer's safety data sheet, the mixture is not classified as hazardous under GHS or the 2012 OSHA Hazard Communication Standard.
