Description

A mouse monoclonal IgG1, kappa antibody from STEMCELL Technologies targeting nestin, a class VI intermediate filament protein expressed by embryonic neuroepithelial stem cells in the developing nervous system and by pancreatic islet and mesenchymal progenitor cells. Clone 10C2 was raised in BALB/c mice against a recombinant protein comprising amino acids 1464 – 1614 of human nestin fused to glutathione S-transferase. The unconjugated reagent reacts with human and cynomolgus nestin and reportedly cross-reacts weakly with monkey nestin but not with rodent nestin.

The antibody is supplied in a phosphate-buffered solution, pH 7.2, containing 0.09% sodium azide and has been purified by affinity chromatography. It has been verified for flow cytometry (FC), immunocytochemistry (ICC), immunofluorescence microscopy (IF) and western blotting (WB), and has been reported for use in immunohistochemistry (IHC) and immunoprecipitation (IP). A doublet band of approximately 220 – 250 kDa is observed on western blots. Suggested working conditions are FC at ≤ 0.2 μg per 1 × 10⁶ cells in 100 μL; ICC/IF at ≤ 0.5 μg/mL; and WB at 0.5 – 2 μg/mL. The product is stable when stored undiluted at 2 – 8 °C and should not be frozen.

Functionally, nestin is required for the survival and renewal of neural progenitor cells, contributes to cellular remodeling through heterodimer formation with vimentin or alpha-internexin, and localizes to growth cones during axon elongation in differentiating neurons. Expression is down-regulated during differentiation and typically disappears by embryonic stage E18. The protein is additionally detected in glioblastomas and in many primary and metastatic melanomas, where it serves as a useful angiogenic marker for evaluating tumor neovascularity.
