Anti-Hu Nestin, 10C2, AD, 25µg

A mouse monoclonal IgG1, kappa antibody against human nestin, offered by STEMCELL Technologies as the Alexa Fluor® 488 conjugate. The 10C2 clone was raised against a recombinant protein comprising amino acids 1464 – 1614 of human nestin fused to glutathione S-transferase. The antibody recognizes a class VI intermediate filament protein expressed by embryonic neuroepithelial stem cells in the developing nervous system and by pancreatic islet and mesenchymal progenitor cells.

Nestin is required for the survival and renewal of neural progenitor cells and plays roles in cellular remodeling by forming heterodimers with vimentin or alpha-internexin, a process that appears to be tightly regulated by phosphorylation. It localizes to growth cones during axon elongation in differentiating neurons, and its expression is down-regulated during differentiation, typically disappearing by embryonic stage E18. The clone reportedly cross-reacts weakly with cynomolgus monkey nestin but not with rodent nestin.

Verified applications are flow cytometry, immunocytochemistry, and immunofluorescence microscopy, with immunohistochemistry reported. On western blots, a doublet band of ~220 – 250 kDa is observed. Suggested use is ≤ 1 μg per 1 x 10^6 cells in 100 μL for flow cytometry and ≤ 10 μg/mL for ICC/IF. The clone has been verified for labeling human neural stem and progenitor cells cultured with STEMdiff™ Neural Induction Medium, STEMdiff™ Neural Progenitor Medium, or NeuroCult™ NS-A Proliferation Kit (Human). The product is supplied in an aqueous buffer containing 0.09% sodium azide, may contain carrier protein/stabilizer, and was purified by column chromatography. Store undiluted at 2 – 8 °C; do not freeze, and protect from prolonged exposure to light.
