Anti-Hu Nestin, 10C2, AD, 100µg

STEMCELL Technologies offers this Alexa Fluor® 488-conjugated mouse monoclonal antibody (clone 10C2, IgG1, kappa) raised against human and cynomolgus nestin. Nestin is a class VI intermediate filament (IF) protein expressed by embryonic neuroepithelial stem cells in the developing nervous system, as well as by pancreatic islet and mesenchymal progenitor cells. Expression is also observed in glioblastomas and many primary and metastatic melanomas, making it a useful angiogenic marker for evaluating neovascularity of endothelial cells in tumors. Nestin is required for the survival and renewal of neural progenitor cells and plays roles in cellular remodeling by forming heterodimers with vimentin or alpha-internexin. Its expression is downregulated during differentiation, typically disappearing by embryonic stage E18.

The 10C2 antibody was generated in BALB/c mice using a recombinant immunogen comprising amino acids 1464–1614 of human nestin fused to glutathione S-transferase, and was purified by column chromatography. It reportedly cross-reacts weakly with monkey nestin but not with rodent nestin. On Western blots, a doublet band of ~220–250 kDa is observed. Verified applications are flow cytometry (FC), immunocytochemistry (ICC), and immunofluorescence microscopy (IF); immunohistochemistry (IHC) has been reported in the literature. Suggested use is ≤ 1 μg per 1 × 10⁶ cells in 100 μL for FC, and ≤ 10 μg/mL for ICC/IF, with titration recommended for optimal performance. The clone has been verified for labeling human neural stem and progenitor cells cultured in STEMCELL Technologies' neural induction and progenitor media.

The product is supplied in an aqueous buffer containing 0.09% sodium azide (may contain carrier protein/stabilizer) and is stable at 2–8°C when stored undiluted. Do not freeze, and protect from prolonged exposure to light. For research use only.
