Anti-Hu Nestin, 10C2, 100µg

STEMCELL Technologies offers a mouse monoclonal IgG1, kappa antibody (Clone 10C2) raised against human nestin, a class VI intermediate filament (IF) protein expressed by embryonic neuroepithelial stem cells in the developing nervous system, as well as by pancreatic islet and mesenchymal progenitor cells. Nestin expression is also observed in glioblastomas and many primary and metastatic melanomas, and it serves as a useful angiogenic marker to evaluate neovascularity of endothelial cells in tumors. The unconjugated antibody was generated using a recombinant protein comprising amino acids 1464 - 1614 of human nestin fused to glutathione S-transferase.

Reactivity is demonstrated with human and cynomolgus nestin, with reported weak cross-reactivity with monkey nestin and no reactivity with rodent nestin. Verified applications include flow cytometry (FC), immunocytochemistry (ICC), immunofluorescence (IF), and Western blotting (WB), with IHC and IP also reported in published studies. On Western blots, a doublet band of approximately 220 - 250 kDa is observed. The clone has been verified for labeling human neural stem and progenitor cells grown with STEMdiff™ Neural Induction Medium, STEMdiff™ Neural Progenitor Medium, and NeuroCult™ NS-A Proliferation Kit (Human).

Suggested use concentrations are: FC, ≤ 0.2 μg per 1 × 10^6 cells in 100 μL volume; ICC/IF, ≤ 0.5 μg/mL; and WB, 0.5 - 2 μg/mL, with titration recommended for optimal performance for each application. The product is supplied as a phosphate-buffered solution at pH 7.2 containing 0.09% sodium azide, and was purified by affinity chromatography. It is stable at 2 - 8 °C when stored undiluted and should not be frozen.
